General description: Ubiquitination: the Notch signaling switch. Recent data reveal that ubiquitination can endorse dual functions in Notch signal transduction pathways. Monoubiquitination and clathrin-dependent endocytosis are required for ?-secretase processing leading to the active form of Notch. We will try to understand the interplay between clathrin-mediated endocytosis of Notch cargo and the monoubiquitination step leading to ligand-mediated receptor activation. The kinetics of these two events will be investigated via the use of different Notch mutants and live-cell imaging techniques. The influence on monoubiquitination and intracellular trafficking of some genetically identified Notch effectors will be studied using siRNAs and dominant-negative mutants.
