NF-kappaB group
General description: Our interest resides is in the signaling events that follow T cell activation through the antigen receptor (TCR). We have shown previously that NEMO, an essential regulator of NF-?B activation is normally recruited to the activated TCR at the immunological synapse. In addition, we have also shown that the presence of NEMO at the cell membrane is sufficient to induce the activation of NF-?B. Our actual goal is to understand the mechanism and consequences of this constitutive activation of NF-?B and importantly to evaluate the status of ubiquitination of NEMO when it is targeted to the membrane. In another project, we have recently shown that ubiquitination targets Bcl10, a component of the TCR-induced NF-?B signaling cascade. This event represents an important negative feed back mechanism of this pathway and our goal is to characterize the molecular events leading to this ubiquitination process.
